Study of the biology of Endothelial Colony Forming Cells (CFCE), a small population that is part of the Endothelial Progenitor Cells considered the true endothelial cells, due to their high potential for proliferation and expansion, as well as their capacity for vascular regeneration in situations of vascular damage, present in umbilical cord blood and peripheral blood of clinically healthy subjects. These cells are used as a control and the functionality and pathophysiology are compared in cells from patients with recurrent deep vein thrombosis. In the same way, I work in the laboratory on the pathophysiology of CFCE, but now in patients recovered from COVID-19 within 30 days and 8 to 12 months of recovery. In the same way, its function is analyzed in aspects related to coagulation in the presence of plasma from patients recovered from COVID-19 as well as plasma from patients who were in moderate to severe condition. For these studies, methodological strategies are used to obtain informed consent from peripheral blood, both from clinically healthy subjects, subjects with recurrent deep vein thrombosis, as well as from patients recovered from COVID-19 at the times described above. Maintenance of cell cultures until CFCE are obtained for subsequent expansion and through molecular biology techniques, the analysis of genes related to endothelial dysfunction, considered as risk factors that could generate thrombosis. In the same way, techniques are used to analyze the expression of proteins in the cell membrane, as well as intracellular and phosphorylated proteins, using multiparametric flow cytometry techniques to determine cellular functionality. On the other hand, tubular structure techniques are used in vitro and in immunodeficient mice to inoculate said human cells and determine if they have angiogenic capacity, form a vascular lumen in the mouse vascular system and determine their function or possible dysfunction.
