PB0739 - Optimization of in vitro and in vivo wound healing effect of platelet-rich plasma

Background: Wound healing is a dynamic process that is responsible for the recruitment and activation of inflammatory cells, angiogenesis, and cellular regeneration. Platelet-rich plasma (PRP) has recently been used in regenerative medicine due to its diverse and abundant growth factors (GFs) and cytokines. Nevertheless, no research has compared PRP's ability to promote wound healing both in vitro and in vivo with the most effective regenerative function.

Aims: We determined to evaluate the wound-healing effect of PRP for optimal use in cellular regeneration.

Methods: We collected canine blood and conducted in vitro and in vivo assays to evaluate the wound-healing effect of PRP.

Results: Whole blood from healthy dogs was centrifuged at various centrifugation conditions, and PRP obtained by centrifugation at 1500 rpm for 10 min showed the highest platelet recovery without other cell contamination. Consistently, platelets obtained from this PRP showed the maximum PDGF-BB release, suggesting that optimizing the centrifugal speed and time is indispensable for the utmost collection of PRP with optimal platelet function. Interestingly, PDGF-BB release was significantly increased upon activation of PRP at 4 hr, indicating the importance of activation time for maximum GF release. Additionally, the group treated with PRP adjusted at 1 × 109 platelets/ml showed significantly enhanced in vitro HUVEC migration, demonstrating the importance of platelet concentration in PRP for better regenerative effect. Furthermore, the activated PRP significantly reduced wound size and improved healing compared to platelet-poor plasma and unactivated PRP in the in vivo rat wound model. Histologically, the skin wound showed significantly accelerated neovascularization, epithelialization, and extracellular matrix formation by PRP, indicating that the application of activated PRP provides the necessary GFs for wound healing.

Conclusion(s): We optimized the in vitro and in vivo wound healing effects of PRP, thereby maximizing the effective and efficient use of PRP in cellular regeneration.
