PB0742 - LYOPHILIZED PLATELET WAFERS (LPCW) COLLAGEN AND CARBOXY METHYLCELLULOSE BASED PLATELET CONCENTRATE PREPARATION – IN VIVO & IN VITRO -PRELIMINARY STUDIES

Background: Till date, autologous platelet concentrates including Platelet Rich Fibrin Matrix( PRFM), PRP Sealants, PRP gels, gelatin PRP impregnated gelatins & sponges, nano fibrin PRP has been attempted whereas only a few studies in platelet wafers have been documented.

Aims: In our ICMR INDIA funded research (2019-21), we aimed at developing Lyophilized Collagen and Carboxy methyl cellulose human and canine platelet wafers as an viable wound healing accelerator for clinical settings.

Methods: LPCW were formulated by freeze-drying to obtain the lyophilized PC wafers as per protocols. In Vitro studies of the LPCW included scanning electron microscopy,, Water loss percent , Gram-Negative Bacterial Endotoxin Assays were studied as per standard protocols. In Vivo Chorioallantoic Membrane (CAM assay), Rabbit pyrogenic testing and wound testing in rabbits (n=6) were studied. Control group-(n=3) wounded and treated with the PC-free wafers, test group (n=3) wounded and treated with the PC wafers and the granulation studied.

Results: In Vitro hydration tests proved the inert swelling properties of the LPCW, water loss results (91.6%), SEM morphology (fig 1) and thus proved the capability to absorb the exudates of the wound. In Vitro Bacterial Endotoxin LAL assay showed minimal bacterial endotoxin levels ( < 0.025 EU/ml). In Vivo Rabbit pyrogenic testing evinced no evident increase in temperature post infusion of reconstituted PCW. In-Vivo wound testing in rabbits, the percentage of wound size reduction during 12days of treatment (from days 0 to 12) (n=9) were documented. The wound granulation and healing was observed within 14 days with human and canine LPCW as compared to as delayed granulation up to 21 days in control rabbits.

Conclusion(s): LPCW were successfully formulated using collagen after various trials and the wafers showed the desired characteristics of the water loss percentage, platelet count, content uniformity, and hydration and hence recommended for preclinical trials for non-healing chronic wounds.
