Background: Previous studies have found significant differences between mammalian embryonic and adult hematopoietic systems, encompassing morphology, molecular characteristics, and functions. However, research on the development-characteristic of platelets, especially the dynamic changes in organelles and the expression of molecules other than classical glycoproteins, is far from comprehensive.

Aims: This study aims to investigate the developmental patterns of platelets on the molecular biological level and explore the correlation between molecular signatures and platelet ultrastructures, as well as functions.

Methods: Platelets enriched from peripheral blood of mice at embryonic (E15.5, embryonic day 15.5), neonatal (P0, postnatal day 0) and adult (8W, 8 weeks) stages were sorted using flow cytometer and subjected to RNA sequencing and protein mass spectrometry. Ultrastructure was studied with transmission electron microscopy. The molecular features and functions of platelets were validated through mito-tracker staining, ATP detection, and granule release test.

Results: In the early stages of individual development, the genes and proteins related to mitochondrial components and ATP synthesis were significantly enriched in platelets. Consistently, organelles comparison demonstrated that platelets in embryo possess a higher content and larger size of mitochondria. Moreover, dense granules, the main storage site for ATP in platelets, were rarely observed in embryonic platelets, while the total amount of ATP released by equal platelets was higher in the embryonic stage, suggesting that embryonic platelets may have a more efficient energy supply system. Conversely, at transcriptomic and proteomic levels, classical coagulation-related molecules increased gradually, reaching their highest expression in adult platelets. Meanwhile, the average volume of α granules, enveloping coagulation factors, gradually increased with development accompanied by a progressively enhanced coagulation response.

Conclusion(s): Omics analysis, ultrastructure observation and corresponding experimental validation indicate that platelets undergo a role transition during mammalian development, potentially evolving from “energy-supplying cells” in the embryonic period to the traditional “procoagulant cells” in the adult stage.
