Background: Ph-negative myeloproliferative neoplasms (MPN) such as polycythemia vera (PV), essential thrombocythemia (ET) and primary myelofibrosis (PMF) are characterized by thrombotic complications. Currently there is not general diagnostic laboratory method for the detection of thrombosis development. Thromboelastography (TEG) is an integral test of hemostasis that evaluates different phases of blood coagulation and the functional state of the hemostasis system. Disorders in the system of fibrinolysis are one of the factors that contribute to the progress of thrombosis. The time of XIIa-dependent fibrinolysis can extend with these patients and represent a change of the balance between blood coagulation and fibrinolysis.

Aims: Aim of this study to estimate parameters of TEG and XIIа-dependent fibrinolysis in patients with MPN.

Methods: The study involved 62 patients with MPN (27 with PV, 14 with ET and 21 with PMF) and 57 healthy controls. It was used thromboelastograph TEG 5000 (Haemonetics, USA). Ly30 and Ly60 were measured. XIIa-dependent fibrinolysis (seconds) was tested by “RENAM” (Russia). STATISTICA 12.0 was used. The results are given as median (Me) with 50 % confidence intervals (CI). Mann-Whitney test was used to compare the studied groups. P< 0.05 was considered statistically significant.

Results: The data was shown in the table.
Ly30 and Ly60 were significantly lower than controls in all patients. Prolongation of time XIIa-dependent fibrinolysis was identified only in patients with PMF.

Conclusion(s): The parameters values indicate the more pronounced fibrinolysis disturbances in patients with PV and PMF. The assay with TEG 5000 more precisely characterizes fibrinolysis disorders than it makes XIIа-dependent method.
