PB0417 - Impact of black vinegar (kurozu) and kurozu moromi powder in mouse models of venous thromboembolism

Background: Venous thromboembolism (VTE) is a general term for deep vein thrombosis (DVT) and pulmonary thromboembolism (PTE) and is increasing over time. Unlike arterial thromboembolism, such as cerebral or myocardial infarction, there have been limited information on foods and their constituents involved in the development and prevention of VTE.

Aims: In this study, we investigated the effects of a pot-brewed pure rice black vinegar (kurozu) concentrate and an insoluble product produced during the fermentation process of kurozu (kurozu moromi powder) in mouse VTE models.

Methods: Kurozu 10-fold concentrate and kurozu moromi powder were provided by Sakamoto Kurozu, Inc. (Kagoshima, Japan). Male C57BL/6J mice were divided into a control diet group, a 0.25% kurozu concentrate-containing diet group, and a 0.5% kurozu moromi powder-containing diet group and maintained for three weeks. DVT was then induced in the mice using electrolysis of a stainless-steel electrode inserted into the inferior vena cava, and thrombus weight in the inferior vena cava was measured two days after treatment. Male C57BL/6J mice were divided into the same three groups and maintained for three weeks. PTE was then induced in mice by intravenous infusion of a collagen + epinephrine mixture that induces platelet aggregation or a tissue factor reagent that activates extrinsic coagulation, and survival and degree of lung vascular occlusion were analyzed.

Results: Both the kurozu concentrate and kurozu moromi powder groups showed a decrease in DVT thrombus weight compared with the control group. Kurozu moromi powder intake reduced collagen + epinephrine-induced PTE symptoms, whereas kurozu concentrate intake did not. Conversely, kurozu concentrate intake improved tissue factor-induced PTE symptoms, whereas kurozu-moromi powder intake did not.

Conclusion(s): Our results suggest that dietary administration of kurozu concentrate and kurozu moromi powder both inhibit venous thrombus formation, but with different mechanisms of action. Therefore, the combination of both may show higher preventive efficacy.
